Transmission electron microscope (TEM) Tecnai G2 Spirit BioTwin from FEI/Thermo Fisher Scientific (www.fei.com/products/tem/tecnai-g2-spirit-for-life-sciences) for the high resolution analysis (magnification up to 300 000x) of the morphology of particles, cells and tissues on ultrastructure level. The system uses 2 CCD cameras (Olympus Veleta ja Gatan Orius) that warrant the sensitivity of detection and enable fast and convenient recording of digitalized images and the software for the image analysis. Microscope also enables acquiring of 3D images of object from semi-thin slices using tilting system (tilt for+/- 80°) and image reconstitution software.
Additionally TEM analysis platform includes ultramicrotome PowerTome MT XL from RMC and Leica’s cryo-ultramicrotome ULTRACUT EM UC7 for slicing the specimens. The latter also enables application of various immuno-electron microscopy techniques. The set of TEM auxiliary equipment also incorporates Leica’s sputter-coater EM ACE600 for preparation of carbon coated grids for TEM analysis. Exactly controlled thickness of deposited carbon layer and glow-discharge of EM ACE600 enable preparation of high quality specimens for analysis of nanoparticles’ morphology.
